London 1868, era of Industrial revolution. Assassin Brotherhood has fallen in London. Jaydeep Mir, A.K.A. henry Green, the son of Indian Assassin Arbaaz Mir, pleaded for help to restore the Assassin brotherhood fearing the worst. He expresses that Templar Grand Master Crawford Starrick, a big player of London's Industry, plans something sinister. Armed with criminals of London in his sleeve, he wants to channelize his wealth & influence to control Great Britain & the world in the process.

In this scenario, Assassin twins Jacob & Evie Frey come to play. They meet Henry Green who introduced himself as an old associate of their father & started hatching plans to counter Templar schemes. Jacob, being a straightforward man is more inclined to the direct fight against the Templars, where as Evie, the more strategic one, suggests they should find the piece of Eden first.

Though they knew none of their plans will work until they free the Templar controlled regions of London. They begin their work by conquering Templar controlled gangs, interfering the main source of Templar wealth...their business & assassinating high ranked officials along with building the team of 'Rooks', their own criminal gang. Frye twins get dignitaries like novelist Charles Dickens, biologist Charles Darwin, inventor Alexander Graham Bell, political thinker Karl Marx, nurse Florence Nightingale, Duleep Singh (the last maharajah of the Sikh Empire), Sergeant Frederick Abberline of the Metropolitan Police Service (known for his investigation on Jack the Ripper), and Queen Victoria as their allies on their war against Templers.

Jacob & start investigation on "Soothing Syrup", a heinous plan of Templar Starrick to slow poison the population of London. He is aided by Charles Darwin. Their investigation leads to a factory which Jacob destroys. He also comes to know about Templar banker Philip Twopenny's plot to steal the gold reserve of Bank of England, which of course he foiled.

Meanwhile, Evie steals an Assassin journal & learns about 'Shroud of Eden', which heals any injury, reverse age & makes the wearer immortal. She manages to locate the vault where this shroud is located & even manages to get the keys, which Templar Throne steals from her. Evie heads for 'Tower of London' & assassinates Throne. In dying breath, Throne informs her that the Shroud is not in the tower at all but located in Buckingham Palace & Crawford Starrick, being an influential member of the society has easy access to the palace.

In the past twins had sets of arguments among themselves where Jacob accused Evie of being inactive & Evie charged Jacob to be irresponsible. Now, twins must lay aside their differences & work as a team to infiltrate Buckingham Palace to obtain 'Shroud of Eden'.
